    Pyronaridine tetraphosphate is a synthetic antimalarial drug belonging to the class of mannich base amino alcohols. It is a tetraphosphate salt of pyronaridine, which enhances its water solubility and stability for pharmaceutical formulation. Its primary and most significant application is as an artemisinin-based combination therapy (ACT) partner drug. Specifically, it is combined with artesunate in the fixed-dose combination therapy known as Pyramax® (pyronaridine-artesunate). This combination is indicated for the treatment of acute, uncomplicated *Plasmodium falciparum* and *Plasmodium vivax* malaria in adults and children. Pyronaridine acts by inhibiting hemozoin formation within the parasite's digestive vacuole, a mechanism similar to that of chloroquine, but it remains effective against many chloroquine-resistant strains. Pyronaridine tetraphosphate is a critical active pharmaceutical ingredient (API) in the global fight against malaria. Its development and use in combination therapies are pivotal for managing drug resistance and improving treatment outcomes in endemic regions.
