    HEPES (4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid) is a zwitterionic organic chemical buffering agent. Its structure features a piperazine ring substituted with a hydroxyethyl group and an ethanesulfonic acid group, giving it a pKa of approximately 7.5 at 25буC. This makes it an excellent buffer in the physiological pH range (6.8-8.2), with minimal metal ion binding and negligible interference with biological reactions. Its primary application is as a non-toxic, non-reactive buffer in cell culture media, particularly for maintaining pH stability in CO_-independent systems. It is also widely used in biochemical and molecular biology assays, including enzyme kinetics, protein purification, and electrophoresis, where consistent pH is critical.
