    2,2-Dihydroxy-1,3-indandione, commonly known as ninhydrin, is a triketone hydrate derivative of indane. It appears as a white to pale yellow crystalline solid and is highly soluble in water and polar organic solvents. Its structure features a fused indane ring system with two geminal hydroxyl groups and a ketone, enabling its characteristic reaction with primary amines. Ninhydrin is primarily utilized as a chromogenic reagent for the detection and quantification of amino acids, peptides, and proteins. Upon reaction with primary amines, it produces a deep purple color known as Ruhemannбпs purple, which is measured spectrophotometrically at around 570 nm. This property makes it indispensable in analytical chemistry for thin-layer chromatography (TLC) visualization, forensic science for latent fingerprint development, and biochemical assays for amino acid analysis. Additionally, ninhydrin serves as a key reagent in clinical diagnostics for monitoring amino acid metabolism disorders and in pharmaceutical research for quality control of amine-containing compounds.
