    4-Nitrophenyl β-D-galactopyranoside is a chromogenic synthetic substrate for β-galactosidase enzymes. It consists of a β-D-galactopyranose moiety linked via a glycosidic bond to a 4-nitrophenol group. This compound is typically supplied as a white to off-white crystalline powder. Its primary and most significant application is as a diagnostic reagent in biochemistry and molecular biology. It is widely used to assay the activity of β-galactosidase (encoded by the *lacZ* gene in *E. coli*). Upon enzymatic hydrolysis by β-galactosidase, the glycosidic bond is cleaved, releasing the yellow-colored 4-nitrophenolate ion, which can be quantified spectrophotometrically at around 405-420 nm. This makes it an essential tool for reporter gene assays, enzyme kinetics studies, and bacterial identification (e.g., in microbiology for differentiating bacterial species based on β-galactosidase activity).
